EVALUATION OF THE EFFECTS OF PRE-INOCULATING CONDITIONS ON THE URINE CULTURE.

Abstract

Aim: To evaluate the role of boric acid as a preservative for urine samples prior to culture and their clinical interpretations by delaying the processing of urine samples up to 3 hours under different storage conditions
Material and Methods: 20 ml of urine was collected in a universal container from 135 patients from the renal medicine and surgical ward of Yangon General Hospital. 10 ml of it was decanted into another container with 0.1 gram of boric acid .From the samples without boric acid 4 aliquots were made in sterile bottles,3 bottles at the room temperature and inoculation on to cysteine lactose electrolyte deficient (CLED) medium was made after 1,2 and 3 hours while 1 aliquot was kept in refrigerator and inoculation was made after 3 hours. The urine preserved with the boric acid was kept at room temperature and inoculation was made after three hours. Result: Out of 135 urine samples 72 sample showed isolation of bacteria and 63 were sterile, it was found that the culture preserved with the boric acid was comparable to those kept at room temperature for 3 hours. After 3 hours at room temperature bacteria count of the urine samples without boric acid was significantly higher than those of the boric acid preservation, Conclusion: The addition of 1% boric acid as preservative is satisfactory for most clinical specimens up to 3 hours after collection especially when rapid transportation to the laboratory or refrigeration is not possible in tropical countries.
