Oxidative stress is a well-known mechanism that is responsible for the development of vascular damage. Minimizing Oxidative stress is an important approach by the use of antioxidants, since antioxidants inhibit the generation of ROS and the subsequent formation of lipid peroxidation products. In the present scenario, a variety of heterocycles have been explored for development of novel antioxidants. From the literature survey 1, 3, 4-oxadiazole was found to be having diverse pharmacological activities such as anti-inflammatory, antimicrobial, antifungal, antiviral, analgesic, anti-mycobacterial, antidepressant and anticancer etc. So it was planned to synthesize a novel series of 1, 3, 4-oxadiazole derivatives and evaluate them for antioxidant activity. Aryl acid hydrazides were synthesized from various aromatic esters, which ultimately react with substituted benzene and succinic anhydride to form aroyl propionic acid. Aroyl propionic acid and aryl acid hydrazide in presence of phosphorus oxychloride to form target 1, 3, 4-oxadiazole derivatives. The synthesized derivatives were authenticated by TLC, UV-Visible, FTIR, NMR and mass spectroscopic techniques. All the target compounds were evaluated for their antioxidant activity by using different in-vitro models. All compounds were found to have good antioxidant activity.
