Cancer is a disease in which the abnormal cells are quite similar to the normal cells, with just minute genetic or functional change. A major disadvantage of anticancer therapy is lack of selectivity for tumor tissue alone, causing severe side effects and resulting in low cure rates. Thus, it is very difficult to target abnormal cells by the conventional method of the drug delivery system. Magnetic microspheres promote drug targeting to tumors with good perfusion. The magnetic microspheres are prepared from albumin, anticancer drug (e.g. doxorubicin) and magnetite. These magnetic microspheres (with size of about 1 micron) are injected as thousands of beads via an incision into the artery - the main blood supply for the organ. When the microspheres are injected, they are picked up by the blood flow and eventually lodge in the malignant cells of the tumors. A magnetic field is then applied to the body to heat the magnetic microspheres and damage the cancer cells. Localization of magnetic spheres in a desired area depends upon magnetic property of microspheres as well as external magnetic field applied to retain it. The magnetic content of microsphere and magnitude of magnetic field applied are crucial -with high magnetic content the magnitude of magnetic field required is decreased but space available for entrapment of drug also decreases. The optimum magnetic content is generally b/w 22%-50% of drug weight in drug carrier complex. The release rate of drug depends upon size and surface area of microspheres; smaller particles have more surface area hence faster release than the larger particles. Chemotherapy is rendered ineffective in case of brain tumors due to impermeable nature of blood brain barrier, magnetic microspheres have been employed as brain tumor targeting system as in case of cationic magnetic aminodextran .Thus magnetic microsphere has been proven as a potential drug delivery system for drug targeting in tumor cells with minimal side effects.
