The process of preparing spherical granules by extrusion/ spheronization was introduced during the late 1960s. It involves forming the powder into a wet mass, which is forced through a restricted area (extrusion) to form strands of extrudate that are broken into short lengths and rounded by placement on a rotating plate within a cylinder. In the production of pharmaceutical pellets, the process of Extrusion & Spheronization is commonly used. Spheronization or merumerization is the process of making extrudes in round shape having diameter of 1mm. Extrusion spheronization follows mainly five steps that is mixing or blending, extrusion, spheronization, coating and finally drying. The result obtained in the spheroniser depends on the rheology of the product. The particles must have enough plasticity to allow deformation under the impact they receive during the spheronization process, but also must be strong enough to withstand the collisions with the friction plate, each other and bowl wall without being broken up and destroyed. Extrusion-spheronization seems to be the most promising process for the optimum delivery of potent drugs having high systemic toxicity. It also offers immense pharmaceutical applicability due to the benefits of high loading capacity of active ingredient(s), narrow size distribution, and cost-effectiveness.
